Mononuclear Phagocytes: Characteristics, Physiology and by S. Gordon, S. Hirsch, P. Starkey (auth.), Ralph van Furth

Sixteen years have handed because the first Leiden convention on Mononuclear Phagocytes. That first assembly resulted in new nomenclature, i. e. , the Mononuclear Phagocytes procedure (MPS), which was once released within the Bulletin of the realm future health association (R. van Furth, et aI. , Bull. WHO, 1972, forty six: 845-852). The paradigm of the MPS has the good thing about being even more constant than the Reticuloendothelial process (RES) (L. Aschoff, Ergebn. lodge. Med. Kinderheilk. 1924, 26:1), simply because cells with related features could be categorized within the comparable class. The essence ota new notion is, in line with Popper, that it needs to be vulnerable to falsification. This has been tried numerous occasions for the MPS by means of those that adhere to the RES. despite the fact that, in biology nobody could be sure of fact. Any concept needs to be in line with experimental observations and/or theoretical contemplate ations and either will be awarded in any such approach that others can try and refute it, then or later. One can have a theoretical or a realistic choice for a concept which at a given time is thought of as very likely truc, yet later it can be proven to be fake. In different phrases, what we found in this quantity is thought of as precise at this second, yet others may possibly reflect on it as now not precise and take a look at to disprove it. Controversies frequently stimulate new research.

Confirmation of the expression of la by renal interstitial cells and other F4/80+ periepithelial cells will require the development of a stable la marker for the mouse and a double labelling method. One population of la-positive cells in the kidney that probably lacks F4/80 is in the glomerulus. la+ -antigen-presenting cells have been localised to the glomerular mesangium (45) but F4/80+ cells are rarely if ever observed in this region in normal adult mice (19). Mesangial cells, which may share some endocytic functions with macrophages, but lack several phenotypic characteristics of the mononuclear phagocyte series (46), also fail to label with F4/80.

CCE was performed following the protocol of Sanderson, et al. (6) in which the rotor speed is maintained at 2500 rpm at a temperature of 18° C. The initial flow rate (driven by a Cole-Parmer Masterflex pump) was 14 mllmin at which most of the platelets were eluted from the system. 0 (22 mllmin). At 22 mllmin, virtually all leukocytes were cleared from the system. The cells from each 100 ml fraction (and an aliquot of the unfractionated population [UF]) were counted to determine yield, and aliquots were resuspended in Minimal Essential Medium containing 10 per cent pooled human AB serum for indirect immunofluorescence analysis and for cytocentrifuge smears.
